PEI, The product name is polyethyleneimine, which is an alkaline cationic polymer widely used in the textile and paper industries. PEI is an alkaline polymer obtained through the polymerization of ethyleneimine. The structural formula CH3CH2N - (- CH2CH2-NH -) n-CH2CH2NH2. The classical n value is 700-2000, and the corresponding polymer molecular weight is 30000-9000Da.
Because the pKa value of amino groups is 10-11, PEI carries a positive charge in neutral pH solutions.
PEI can be considered as soluble DEAE cellulose. PEI can bind to negatively charged macromolecules such as nucleic acids and acidic proteins, forming a network structure of PEI. Subsequently, the bound acidic molecules quickly precipitate. Whether acidic proteins can bind to PEI depends on the concentration of the salt solution. At lower salt concentrations (0.1 mol/L NaCl), slightly acidic proteins will bind to PEI to form a precipitate,
But at moderate salt concentrations (0.4mol/L NaCl), it can be eluted from the polymer and become soluble. Strong acidic proteins bind to PEI at low salt concentrations, but do not dissolve at moderate concentrations. They can only be eluted at high salt concentrations (0.9mol/L NaCl).
It should be noted that when the protein is washed off the PEI particles, both the protein and PEI itself become soluble. So before returning to low salt concentration, PEI needs to be removed from the protein.
